Cobetter TFF Cassettes: Applications and Case Studies in Synthetic Peptide Processing

2026.10.15 54

Peptides are high-value bioactive molecules built from amino acids via peptide bonds, known for their targeting precision and low toxicity. Most peptides are produced via solid-phase synthesis, generating complex crude feed steams that must be purified by chromatography and filtration.

Ultrafiltration/Diafiltration (UF/DF) is a critical step in peptide purification, enabling efficient concentration, removal of low-molecular-weight impurities and solvent.

Key Strengths of Cobetter RC Membranes

Enhanced Organic Solvent Compatibility

Peptide purification processes often involve organic solvents such as acetonitrile and DMSO, requiring membranes with robust chemical compatibility.

While conventional PES membranes offer high flux, their compatibility with certain organic solvents may limit their application in solvent-rich processes.

Cobetter's Regenerated Cellulose (RC) TFF cassettes are designed to address these challenges, offering validated solvent compatibility to support reliable performance in demanding peptide purification applications.

Minimizing product loss is essential in high-value peptide purification

Cobetter's Regenerated Cellulose (RC) membranes exhibit low non-specific protein adsorption (<0.5 μg/cm²), compared with approximately 10 μg/cm² for conventional PES membranes, helping minimize product loss and support higher recovery yields.

Wide Screens for Challenging Feed Streams

Peptide feed streams are characterized by high concentration and viscosity. Cobetter RC TFF cassettes feature on open screen, which is suitable for fluid dynamics of such complex feed streams, thereby minimizing screen blockage and maintaining filtration flux.

Cobetter's RC TFF cassettes feature an open-channel screen design that minimizes flow obstruction and supports stable filtration flux, even under demanding processing conditions.

For solvent-free applications, Cobetter's PES TFF cassettes offer an alternative with high-flux performance, enabling membrane selection based on specific process requirements.

Selection Guide for RC TFF Cassettes at 1~3 kDa MWCO

Synthetic peptides generally exhibit molecular weights between 1,000 and 5,000 Da, in which the conventional UF retention principle of selecting a molecular weight cut-off (MWCO) at 1/3–1/5 of the target molecular weight is not always applicable in peptide processing.

Cobetter recommends the following membrane selection guidelines to support process development.

These recommendations serve as general guidelines. Final process parameters should be optimized and validated based on the specific feed characteristics and process requirements.

Case studies

The following case studies are based on real-world application data and have been anonymized. All data were generated using Cobetter RC TFF cassette with 2 kDa MWCO at a feed flow rate of 5 L/min/m².

Case 1: Acetonitrile Removal

Background

Peptide: ~4.7 kDa, feed stream containing 30% acetonitrile.

Goal: Concentrate and exchange into purified water and remove acetonitrile.

Setup: Cobetter RC TFF cassette at 2 kDa MWCO, TMP at 1.3 bar.

Data from lab-scale testing

Results

Concentration: 2.0× concentration factor (CF), average flux at 5.8 LMH.

Diafiltration: 7.0× diafiltration volume (DV), average flux at 6.7 LMH.

Acetonitrile Removal: At 5 DV, acetonitrile ratio dropped to 0.2093%; at 7 DV, down to 0.0089%.

This case confirms the robust performance of RC TFF cassettes in acetonitrile-containing feed streams and their efficient solvent exchange. At 7 DV, acetonitrile residue dropped to a negligible level, well within downstream process requirement.

Case 2: Challenges of High CF & High Final Concentration

Background

Peptide: 3–4 kDa.

Process: Concentration → diafiltration → over-concentration, demanding stable flux and strong concentration capability.

Setup: Cobetter RC TFF cassette at 2 kDa MWCO, TMP at 2.5 bar.

Data from lab-scale-testing

Results

Concentration Ⅰ: 11.0× CF, average flux at 12 LMH.

Diafiltration: 4.5× DV, average flux at 12 LMH.

Concentration Ⅱ: Further 2.0× CF, average flux at 5.8 LMH.

Outcome: 96.6% product yield; retentate concentration reached 97 mg/mL.

This case highlights the stable flux and excellent yield of RC TFF cassettes across the full process. Through 11× concentration, 4.5× diafiltration, and 2× over-concentration, the cassette delivered 96.6% yield and a final concentration of 97 mg/mL—proving its reliability under tough conditions.

Case 3: High Yield of >99%

Background

Peptide: ~4.3 kDa.

Process: 10× CF following 8× DV.

Setup: Cobetter RC TFF cassette at 2 kDa MWCO, TMP at 2.0 bar.

Data from lab-scale-testing

Results

Concentration: 10.0× CF, average flux at 12.6 LMH.

Diafiltration: 8.0× DV, average flux at 10.7 LMH.

Concentration Ⅱ: Further 2.0× CF, av.erage flux at 5.8 LMH.

Yield: >99%.

This case demonstrates the potential of the RC TFF cassettes in achieving exceptionally high recovery yields. For high-value peptide products, every percentage increase in yield translates into significant economic benefits.
